Background

Pro Caspase-1 is a 45 kDa inactive zymogen that, upon assembly of the canonical inflammasome, undergoes autoproteolysis between the p20 and p10 subunits, releasing an N-terminal CARD domain, the p20 subunit, and the p10 subunit; the resulting heterotetramer of two p20 and two p10 chains (often described as p20-p10 plus the additional p12 peptide that can remain non-covalently associated) forms the active caspase-1 protease, which subsequently cleaves pro-IL-1β and pro-IL-18 to drive inflammatory responses.
