Background

PPIF/Cyclophilin F protein is a member of the peptidyl-prolyl cis-trans isomerase (PPIase) family, which catalyzes the isomerization of proline imide peptide bonds in oligopeptides, thereby accelerating protein folding. It is a critical component of the mitochondrial permeability transition pore (mPTP) in the inner mitochondrial membrane, and its activation is associated with the induction of apoptotic and necrotic cell death. Cyclophilin F also interacts with mitochondrial p53/TP53 to activate necrosis induced by oxidative stress and regulates mitochondrial membrane F(1)F(0) ATP synthase activity. Additionally, it has anti-apoptotic activity independent of mPTP, cooperating with BCL2 to inhibit cytochrome c-dependent apoptosis.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human colon. Anti-PPIF/Cyclophilin F antibody was used at 1/200 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry

ICC shows positive staining in HepG2 cells. Anti-PPIF/Cyclophilin F antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
