Background
PPIA (peptidyl-prolyl cis-trans isomerase A), also called cyclophilin A, is a 165-amino-acid, ubiquitously expressed cytoplasmic enzyme that catalyzes the cis-trans isomerization of prolyl peptide bonds and thus accelerates protein folding; beyond this chaperone-like role, it is a critical virulence factor for HIV-1, because the viral capsid recruits PPIA to shield itself from host restriction factors, while extracellular PPIA, secreted under inflammatory or oxidative stress, acts as a chemoattractant for monocytes and T cells via CD147/EMMPRIN, thereby amplifying inflammatory cascades in cardiovascular disease, viral infection, and cancer.

Flow cytometric analysis of K562 (Human chronic myelogenous leukemia lymphoblast) labelling PPIA antibody at 1/500 dilution (0.1 μg) / (Red) compared with a Rabbit monoclonal IgG (Black) isotype control and an unlabelled control (cells without incubation with primary antibody and secondary antibody) (Blue). Goat Anti - Rabbit IgG Alexa Fluor® 488 was used as the secondary antibody.

Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
