Background

Phospho-serine and phospho-threonine are crucial post-translational modifications where a phosphate group is covalently attached to the hydroxyl side chain of specific serine or threonine residues within a protein. This phosphorylation event, primarily catalyzed by enzymes known as kinases, dramatically alters the protein's properties by introducing a bulky, negatively charged group. Consequently, it can induce conformational changes, create new binding sites for partner proteins, and fundamentally regulate the protein's activity, stability, localization, and interactions with other biomolecules. As one of the most ubiquitous and essential regulatory mechanisms in biology, serine/threonine phosphorylation is a central control point in countless cellular processes, including signal transduction, cell cycle progression, gene expression, and apoptosis. The dynamic nature of this modification, which can be reversed by phosphatases, allows cells to rapidly and reversibly respond to both internal and external stimuli.
