Background
Phospho-RIP (Ser166) is the phosphorylated form of receptor-interacting protein kinase 1 (RIPK1) at serine 166, a modification that acts as a biomarker for the kinase’s activation and marks the initiation of RIPK3/MLKL-dependent necroptotic signaling; this phosphorylation event converts RIPK1 from a pro-survival scaffold into a death-inducing kinase, enabling assembly of the necrosome complex (RIPK1–RIPK3–MLKL) that drives inflammatory, caspase-independent cell death, and its detection is therefore widely used to monitor necroptotic activity in cancer, neurodegeneration and inflammatory disease research.
