Background

Phospho-PKA Substrate (RRXST) is a recognition motif for substrates specifically phosphorylated by protein kinase A (PKA). Within this motif, serine (S) or threonine (T) serves as the phosphorylation site, flanked on its N-terminal side by two arginine residues (RR). The position immediately following the second arginine can be any amino acid (X). This highly conserved pattern of basic amino acids ensures that PKA can specifically recognize and catalyze the phosphorylation of serine/threonine residues in its substrates, thereby regulating various critical cellular signaling pathways, including glycogen metabolism, gene transcription, and cell proliferation. Specific phospho-antibodies targeting this motif have become essential research tools for detecting PKA activity and its downstream signaling events.
