Background
Phospho-PDGF Receptor α (Tyr1018) is the activated form of the platelet-derived growth factor receptor alpha (PDGFRα) following phosphorylation at tyrosine residue 1018, representing the activation state of this receptor tyrosine kinase signaling pathway. This site is located within the kinase insert region of PDGFRα. Upon ligand binding (such as PDGF-AA, PDGF-BB, or PDGF-CC), the receptor undergoes dimerization and autophosphorylation, with phosphorylation at Tyr1018 providing a critical docking site for the signaling adaptors Grb2 and Shc, thereby activating the Ras-MAPK pathway to regulate cell proliferation, survival, and migration. Studies have shown that aberrant phosphorylation at Tyr1018 is closely associated with sustained activation of PDGFRα in conditions such as idiopathic pulmonary fibrosis, atherosclerosis, and various tumors (e.g., glioblastoma, gastrointestinal stromal tumors).
