Background
Phospho-IRAK4 (Thr345/Ser346) is the autophosphorylated, catalytically active form of interleukin-1 receptor-associated kinase 4 that forms within minutes of TLR/IL-1R stimulation; these two adjacent residues in the activation loop must be phosphorylated—chiefly through IRAK4 trans-autophosphorylation after MyD88-mediated dimerization—to stabilize the loop, fully unleash kinase activity toward IRAK1 and Pellino E3 ligases, and thereby propagate downstream NF-κB and MAPK inflammatory signaling.
