Background
PEX14, encoded by the PEX14 gene, is an integral peroxisomal membrane protein whose C-terminus faces the cytosol and constitutes the docking site for the PTS1 receptor PEX5 (and, indirectly, the PTS2 receptor PEX7), thereby nucleating the PEX13-PEX14 translocon pore that imports peroxisomal matrix proteins; beyond its essential role in peroxisome biogenesis, PEX14 also binds microtubules to drive organelle motility, functions as a transcriptional corepressor by recruiting histone deacetylases, and—uniquely among peroxins—is strictly required for selective autophagic degradation of peroxisomes (pexophagy), while mutations in PEX14 produce a Zellweger-syndrome phenotype .

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human testis. Anti-Pex14 antibody was used at 1/100 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry
ICC shows positive staining in HepG2 cells. Anti-Pex14 antibody was used at 1/50 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
