Background
p35/25 is a neuron-specific activator of cyclin-dependent kinase 5 (CDK5): p35 is the full-length, membrane-anchored protein that rapidly turns over and keeps CDK5 activity tightly controlled, whereas p25 is the calpain-cleaved, N-terminally truncated fragment that lacks the membrane-targeting myristoylation signal, relocates CDK5 to nuclear/perinuclear compartments, and—because of its 5- to 10-fold longer half-life—induces prolonged, deregulated CDK5 hyperactivity that aberrantly phosphorylates tau and other cytoskeletal proteins, thereby contributing to neurodegeneration in Alzheimer’s disease and related tauopathies.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human cerebral cortex. Anti-p35/25 antibody was used at 1/250 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Heat mediated antigen retrieval with EDTA buffer pH9.0 was performed before commencing with IF staining protocol.
