Background
NLRP3 (NOD-, LRR- and pyrin domain-containing protein 3) is a ~118 kDa cytosolic pattern-recognition receptor that assembles into the macromolecular “NLRP3 inflammasome” when confronted by a structurally diverse set of microbial toxins, RNA viruses, extracellular ATP, crystalline substances or metabolic danger signals; upon activation NLRP3 nucleates the adaptor ASC and pro-caspase-1 filaments, driving caspase-1–mediated proteolytic maturation of interleukin-1β/18 and gasdermin D, which triggers pyroptotic cell death and amplifies systemic inflammation, thereby placing NLRP3 at the hub of innate immunity, autoinflammatory syndromes and chronic metabolic disorders such as gout, atherosclerosis, Alzheimer’s disease and type-2 diabetes.
