Background
MRP2 (Multidrug Resistance-associated Protein 2), also known as ABCC2, is a 1,545-amino-acid ATP-binding cassette (ABC) transporter localized almost exclusively to the apical (canalicular) membrane of hepatocytes, where it functions as the primary efflux pump for conjugated organic anions including bilirubin glucuronides, glutathione conjugates, and a wide array of xenobiotics such as anticancer drugs, antibiotics, and environmental toxins, thereby playing a central role in hepatic detoxification and limiting the systemic exposure of tissues to these compounds; genetic mutations in the ABCC2 gene lead to Dubin–Johnson syndrome, a benign autosomal recessive disorder characterized by conjugated hyperbilirubinemia and black liver, whereas acquired MRP2 down-regulation contributes to cholestasis and multidrug resistance in cancer chemotherapy, and its expression is transcriptionally regulated by nuclear receptors like PXR, CAR, and FXR that respond to endobiotics and xenobiotics, making MRP2 a critical determinant of pharmacokinetics, drug–drug interactions, and bile formation.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human kidney. Anti-MRP2 antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
