Background

Mouse RBP4 (Retinol-Binding Protein 4) is a 21-kDa lipocalin-family protein featuring a conserved β-barrel structure that encapsulates all-trans retinol within its hydrophobic core. It mediates systemic vitamin A transport by forming a stabilizing complex with transthyretin (TTR), thereby extending its plasma half-life and preventing renal clearance. The mouse ortholog shares >90% sequence identity with human RBP4, including critical retinol-binding residues (e.g., Leu39, Phe61), but exhibits distinct kinetics in TTR binding compared to rat and human variants. Functionally, RBP4 ensures retinoid delivery to peripheral tissues, supporting vision, embryogenesis, and epithelial maintenance.
