Background

CD45R, also known as B220, is a protein tyrosine phosphatase that is predominantly found on B cells, at varying levels from pro-B cells to activated B cells, and may also be detected on certain T cell and NK cell subsets. CD45R plays a crucial role in B cell receptor and T cell receptor signaling and is essential for lymphocyte development and function. It helps regulate the threshold of sensitivity to external stimuli, affecting cellular responses. In mice, CD45R/B220 is a common marker for B cells, whereas in humans, B cells do not express B220 in the same way; it appears to be differentiation-specific and is therefore expressed on only some B cell subsets. CD45R is also used in the classification of leukemias and lymphomas and in research on functional subsets of B and T cells.

Flow cytometric analysis of C57BL/6 mouse splenocytes labeling Mouse CD45R/B220 at 1/2000 (0.1 μg) dilution / (Right panel) compared with a Rat IgG, Isotype Control / (Left panel). Goat Anti - Rat IgG Alexa Fluor® 488 was used as the secondary antibody. Then cells were stained with CD3 - Brilliant Violet 421™ separately. Gated on total viable cells.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded mouse spleen. Anti-Mouse CD45R/B220 antibody was used at 1/500 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Negative control: IHC shows negative staining in paraffin-embedded mouse cerebral cortex. Anti-Mouse CD45R/B220 antibody was used at 1/500 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunofluorescence

IF shows positive staining in paraffin-embedded mouse spleen. Anti-Mouse CD45R/B220 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rat IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. Counterstained with DAPI (Blue). Heat mediated antigen retrieval with EDTA buffer pH9.0 was performed before commencing with IF staining protocol.
