Background
Matrix metalloproteinase-14 (MMP14, MT1-MMP) is a trans-membrane, zinc-dependent endopeptidase that serves as a master regulator of pericellular proteolysis by cleaving virtually all extracellular-matrix proteins (collagens, laminins, fibronectin, proteoglycans), activating pro-MMP2 and pro-MMP13, shedding cell-surface receptors and adhesion molecules, and thereby governing development, angiogenesis, wound repair, adipogenesis and alveolarization, while its chronic over-expression—driven by TGF-β/SMAD, HIF-2α/Sp1 or EGF signalling and restrained by TIMP-2, microRNAs and promoter methylation—promotes tumour invasion, metastasis, fibrosis and obesity, making it a key therapeutic target in cancer, Graves’ orbitopathy, BPD and other ECM-related disorders.
