Background
LY6E/SCA-2 is a ~14 kDa glycosylphosphatidylinositol-anchored cell-surface protein of the Ly6/uPAR superfamily that is constitutively expressed in thymus, spleen, placenta and many epithelia and is strongly up-regulated by type I interferons; it functions as a context-dependent modulator of immunity and oncogenesis by tuning T-cell receptor/CD3ζ phosphorylation, enhancing HIV-1 and other viral replication while conversely restricting coronavirus entry via spike-protein interference, and its over-expression in multiple tumors promotes immune escape, metastasis and chemoresistance, making it a candidate prognostic marker and therapeutic target.

Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry
ICC shows positive staining in T-47D cells. Anti-LY6E/SCA-2 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
