Background
Latent Membrane Protein 2A (LMP2A) is a viral protein encoded by the Epstein-Barr virus (EBV) and is expressed during the latent phase of EBV infection in B cells. It is a transmembrane protein with three distinct domains: an N-terminal cytoplasmic domain containing ITAM motifs that mimic B-cell receptor (BCR) signaling, a transmembrane domain with 12 segments, and a C-terminal domain that mediates protein dimerization. LMP2A plays a crucial role in maintaining viral latency by blocking BCR signal transduction, thereby preventing B-cell activation and apoptosis. Additionally, it has been implicated in promoting B-cell survival and proliferation through pathways such as the mTOR pathway and by modulating cellular metabolism. LMP2A is consistently expressed in various EBV-associated malignancies, including Burkitt's lymphoma, Hodgkin's lymphoma, and nasopharyngeal carcinoma, highlighting its significance in viral oncogenesis.
