Background
LIN28B is an evolutionarily-conserved RNA-binding protein that acts as a potent developmental timing regulator and oncofetal factor by selectively binding and blocking the biogenesis of let-7 family microRNAs, thereby relieving repression of pluripotency, cell-cycle, and metabolic genes; it is predominantly expressed during embryogenesis and in many cancers, where its overexpression promotes self-renewal, metabolic reprogramming, and aggressive phenotypes through both let-7–dependent and independent mechanisms, including direct stabilization of IGF2, HMGA1, and MYC mRNAs and activation of the PI3K/AKT/mTOR and WNT/β-catenin pathways, making LIN28B a critical driver of oncogenesis, stemness, and resistance to therapy across diverse tumor types.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human placenta. Anti-LIN28B antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry
ICC shows positive staining in K562 cells. Anti-LIN28B antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
