Background
KLK6 (kallikrein-related peptidase 6) is a secreted serine protease encoded on chromosome 19q13.3–13.4 that exerts context-dependent regulation of tumor behavior by cleaving extracellular matrix proteins and activating protease-activated receptors, thereby promoting proliferation, migration, invasion, angiogenesis, and chemoresistance in gastric, colorectal, lung, bladder, and other cancers while also modulating epithelial-to-mesenchymal transition and macrophage–tumor crosstalk via PAR1/PAR2 signaling cascades, making its elevated expression a consistent biomarker of poor prognosis.
