Background

Insulin-like growth factor-binding protein 7 (IGFBP7), also known as MAC25 or prostacyclin-stimulating factor, is a secreted glycoprotein belonging to the IGFBP superfamily that uniquely exhibits low affinity for insulin-like growth factors (IGFs) compared to other family members, thereby functioning primarily through IGF-independent mechanisms to regulate critical cellular processes such as senescence, apoptosis, and angiogenesis. Originally identified as a tumor suppressor frequently downregulated or silenced via promoter hypermethylation in various malignancies including breast, colorectal, and prostate cancers, IGFBP7 exerts its anti-tumor effects by inhibiting the RAS-MAPK signaling pathway and disrupting endothelial cell tube formation, while also playing a pivotal physiological role in vascular biology by stimulating prostacyclin production and maintaining vascular tone. Beyond its oncological significance, emerging research highlights its involvement in metabolic regulation, kidney injury responses, and fibrotic diseases, positioning IGFBP7 not only as a promising diagnostic and prognostic biomarker but also as a potential therapeutic target for restoring normal cell cycle control and inhibiting pathological neovascularization.

Immunocytochemistry

ICC shows positive staining in SK-OV-3 cells. Anti-IGFBP7 antibody was used at 1/100 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
