Background

IFT88 (intraflagellar transport protein 88) is an 833-amino-acid tetratricopeptide-repeat protein that serves as a core subunit of the IFT-B complex, powering anterograde delivery of ciliary cargo by kinesin motors, and is therefore indispensable for assembly and maintenance of motile and primary cilia; beyond this canonical ciliary role, IFT88 also localizes to centrosomes in proliferating non-ciliated cells where it binds the Rb regulator Che-1 to restrain G1-S transition, acts in mitotic spindle orientation, endocytic recycling of immune receptors, angiogenesis, and cartilage homeostasis, while mutations or depletion of IFT88 produce developmental malformations, polycystic kidney disease, skeletal defects, retinal degeneration, and tumor progression, illustrating its broad importance in both cilium-dependent and cilium-independent cellular processes.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human testis. Anti-IFT88 antibody was used at 1/200 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
