Background
HOPX (homeodomain-only protein) is a 73-amino-acid transcriptional co-factor that contains an atypical homeodomain lacking residues required for DNA binding; instead, it modulates gene expression by interacting with partners such as serum response factor (SRF) to regulate cardiac development, stem-cell fate, and differentiation in multiple tissues. Expressed in embryonic and adult stem cells (e.g., hippocampal neural stem cells), HOPX acts as a tumor suppressor whose loss or silencing is common in cancers (e.g., choriocarcinoma, lung, head and neck squamous carcinoma, and esophageal cancer) and is implicated in regulating immune cell homeostasis, including regulatory T cells and effector T-cell differentiation.
