Background

Histone H2B (mono methyl K5) is a specific modification of the histone protein H2B at lysine residue 5, where a single methyl group is added. Histone H2B (mono methyl K5) is a core component of the nucleosome, which is essential for the packaging of DNA into chromatin. This modification, along with others, contributes to the regulation of DNA accessibility, thereby influencing transcription regulation, DNA repair, DNA replication, and chromosomal stability. Histone H2B mono methylation, particularly at K5, has been implicated in transcriptional regulation. It is part of a complex set of post-translational modifications known as the "histone code," which affects gene expression.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human kidney. Anti-Histone H2B (mono methyl K5) antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Anti-Histone H2B (mono methyl K5) antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).

ICC shows positive staining in NIH/3T3 cells. Anti- Histone H2B (mono methyl K5) antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
