Background
GORASP2 (also called GRASP55) is a peripheral membrane protein encoded by the GORASP2 gene, located on the medial-trans face of the Golgi apparatus, where it acts as a "molecular glue" by forming trans-oligomers through its conserved N-terminal GRASP domain to stack adjacent cisternae into the characteristic Golgi ribbon . In normal growth conditions it is O-GlcNAcylated, but upon glucose or energy starvation it is rapidly de-O-GlcNAcylated and a subpopulation relocates to the autophagosome–lysosome interface, where it tethers LC3-II on autophagosomes to LAMP2 on lysosomes, thereby facilitating autophagosome–lysosome fusion and promoting autophagosome maturation as an unconventional energy sensor.

Immunocytochemistry
ICC shows positive staining in HeLa cells. Anti- GORASP2 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
