Background

The green fluorescent protein (GFP) is a ~27 kDa protein composed of 238 amino acids, originally isolated from the jellyfish Aequorea victoria. Its core structure features a unique β-barrel fold with a central fluorescent chromophore (formed by the autocatalytic cyclization and oxidation of Ser65-Tyr66-Gly67). When excited by blue light at 395 nm (primary peak) or 475 nm (secondary peak), GFP emits green fluorescence at 509 nm without requiring additional cofactors. This property has made GFP a revolutionary tool in molecular and cellular biology, widely used for gene expression reporting, protein localization tracking, and dynamic cell imaging.

GFP transfected 293T (Human embryonic kidney epithelial cell (Right panel) or 293T (Left panel) was stained with Alexa Fluor® 647 Rabbit IgG Isotype Control (Black line histogram) and SDT GFP Recombinant Rabbit mAb (Alexa Fluor® 647 Conjugate) (Red line histogram) at 1/1000 dilution (0.1 μg), cells without incubation with primary antibody and secondary antibody (Blue line histogram) was used as unlabelled control. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.

Immunocytochemistry

ICC shows positive staining in Histone H3-GFP transfected 293F cells. Anti- GFP (Alexa Fluor® 647 Conjugate) antibody was used at 1/50 dilution (meganta) and incubated overnight at 4°C. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue).

ICC shows negative staining in vector-transfected 293F cells. Anti- GFP (Alexa Fluor® 647 Conjugate) antibody was used at 1/50 dilution and incubated overnight at 4°C. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue).
