Background
GBP5 (Guanylate-Binding Protein 5) is a ~67 kDa interferon-inducible large GTPase that polymerizes on membranes and cytosolic surfaces to exert antimicrobial and inflammatory signaling functions; upon pathogen recognition it catalyzes GTP hydrolysis, oligomerizes into coat-like structures that destabilize bacterial or parasitophorous vacuole membranes, recruits the NLRP3 inflammasome via direct CARD-CARD interaction to drive caspase-1–dependent IL-1β/IL-18 maturation, and modulates autophagy and pyroptosis, thereby acting as a critical cytosolic pattern recognition receptor that links innate immunity to cell-autonomous defense against bacteria, viruses, and protozoa while its dysregulation contributes to autoinflammatory diseases and cancer.
