Background

GABRA1 (gamma-aminobutyric acid type A receptor subunit alpha1) is the most abundant α-subunit of the pentameric GABA_A receptor, the brain’s principal fast inhibitory ion channel, and its TM2 helix lines the chloride-selective pore; mutations like A322D or E177A reduce surface expression or gating, causing severe epileptic encephalopathy, while single-nucleotide polymorphisms modulate sensitivity to benzodiazepines, ethanol and general anesthetics that bind at the α1/γ2 interface, making GABRA1 a pharmacological cornerstone for sedation, sleep and anesthesia as well as a high-priority target for precision therapy of genetic epilepsy.

IHC shows positive staining in paraffin-embedded human cerebral cortex. Anti-GABRA1 antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Negative control: IHC shows negative staining in paraffin-embedded human colon. Anti-GABRA1 antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
