Background

FTH1, also known as Ferritin Heavy Chain 1, is a protein-coding gene that plays a crucial role in iron metabolism. It encodes a subunit of ferritin, the major intracellular iron storage protein in both prokaryotic and eukaryotic organisms. The ferritin protein is composed of 24 subunits of heavy and light chains, with FTH1 being the heavy chain component. This protein helps catalyze the oxidation of Fe²⁺ to Fe³⁺, facilitating iron storage in a soluble and non-toxic form. Additionally, FTH1 possesses ferrous iron binding activity and is involved in various biological processes, including immune response, cell proliferation, and protection against oxidative stress. Defects in the FTH1 gene are associated with several diseases, such as neurodegenerative disorders and iron overload conditions like Hemochromatosis Type 5.

Immunocytochemistry

ICC shows positive staining in Jurkat cells. Anti- FTH1 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
