Background

DMC1, a meiosis-specific RecA-homologous recombinase conserved from fungi to humans, is indispensable for accurate homologous chromosome segregation during meiosis: it loads onto the single-stranded DNA tails generated at programmed DNA-double-strand breaks, forms octameric rings that encircle the DNA, and drives the search for and strand invasion of homologous chromatids, thereby catalyzing the crossover events that stabilize bivalents; in so doing it works alongside (and partly suppresses) the ubiquitous RAD51, and its absence or mutation causes meiotic arrest, defective synaptonemal-complex formation, and sterility .

Immunohistochemistry

IHC shows positive staining in paraffin-embedded mouse testis. Anti-DMC1 antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Negative control: IHC shows negative staining in paraffin-embedded mouse liver.
