Background

DLX5 is a distal-less homeobox transcription factor essential for embryonic development, particularly in osteogenesis, chondrogenesis, and craniofacial patterning, where it activates Runx2 and Sp7 to drive osteoblast differentiation while also regulating chondrocyte hypertrophy and coupling bone formation to resorption by modulating the RANKL/OPG ratio in mesenchymal cells; it binds the 5'-TAATTA-3' consensus, synergizes with BMP signaling, and its human mutations are linked to split-hand/split-foot malformation, sensorineural hearing loss, and enhanced osteosarcoma progression via NOTCH pathway activation.

The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
