Background

Cytochrome P450 proteins constitute a ubiquitous superfamily of heme-containing mono-oxygenases that orchestrate the regio- and stereoselective insertion of a single oxygen atom into thousands of structurally diverse substrates, leveraging a conserved Fe(II)-protoporphyrin IX center that cycles between ferric, ferrous, and highly reactive Fe(IV)=O (Compound I) states to catalyze reactions ranging from hydroxylation and epoxidation to dealkylation and dehalogenation, thereby governing the phase-I metabolism of approximately 75 % of all pharmaceuticals, detoxifying xenobiotics, synthesizing endogenous signaling molecules such as steroid hormones and eicosanoids, and driving biosynthetic pathways in plants, fungi, and bacteria, while exhibiting remarkable inter-species and inter-individual genetic polymorphisms that underlie variable drug responses, environmental adaptations, and evolutionary diversification across archaea, prokaryotes, and all eukaryotic kingdoms.
