Background
Bovine RHOXF1 is a ~20.5 kDa reproductive-homeobox transcription factor that shuttles between nucleus and cytoplasm in testicular and mammary cells, where it preferentially binds promoters of fertility-related and metabolic genes—such as ACADVL, IRF6, APOA5 and AKT3—and generally represses their transcription, thereby modulating lipid catabolism, β-casein synthesis and milk-component secretion; in cattle populations, natural SNPs that disrupt its DNA-binding motif are associated with significant differences in milk-fat and protein yield, while in vitro studies indicate that RHOXF1 silencing accelerates apoptosis and reduces proliferation of bovine mammary epithelial cells, collectively pointing to a central, dose-sensitive role in coordinating male germ-cell maturation with lactation physiology.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded bovine testis. Anti-Bovine RHOXF1 antibody was used at 1/500 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
