Background

Anterior Gradient 2 (AGR2) is an ~20 kDa endoplasmic-reticulum–resident protein disulfide-isomerase that catalyzes disulfide-bond formation in nascent mucins and other secreted proteins, acts as a molecular chaperone to maintain ER proteostasis, and is highly expressed in mucosal epithelia; in cancers its overexpression is linked to aggressive growth, metastasis, chemoresistance and suppression of type-I interferon responses, while its loss triggers ER stress and the unfolded-protein response, and a secreted monomeric form detectable in serum and urine serves as a promising extracellular biomarker.

Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
