Background

ALOX5, also known as 5-lipoxygenase, is a key enzyme involved in the metabolism of arachidonic acid, a polyunsaturated fatty acid. It catalyzes the conversion of arachidonic acid to leukotriene A4, which is then further metabolized into various leukotrienes such as leukotriene B4 and cysteinyl leukotrienes. These leukotrienes play significant roles in inflammatory and allergic responses, including bronchoconstriction, increased vascular permeability, and chemotaxis of immune cells. ALOX5 is primarily expressed in immune cells like neutrophils, macrophages, and mast cells. Its activity is tightly regulated at multiple levels, including transcriptional control, post-translational modifications, and interaction with other proteins. Inhibitors of ALOX5 have been developed as potential therapeutic agents for conditions like asthma, allergic rhinitis, and certain inflammatory diseases, aiming to reduce the production of pro-inflammatory leukotrienes and alleviate associated symptoms.
