Background
LysoTracker Green DND-26 is a cell-permeant, non-fixable green fluorescent dye specifically designed for staining acidic compartments within cells, such as lysosomes. It has an optimal excitation/emission wavelength of 504/511 nm and can be effectively excited using a FITC filter.
Simple, Highly Specific One-Step Staining and Tracking of Acidic Organelles
The LysoTracker probe consists of a fluorescent group combined with a weak base, which is only partially protonated under neutral pH conditions. In its neutral charge state, the probe can freely diffuse across the intact plasma membrane of live cells. When the probe diffuses into lysosomes, the weak basic group becomes protonated due to the acidic environment inside the lysosomes. In this charged state, the probe is less likely to diffuse across the organelle membrane, leading to local accumulation within acidic organelles (such as lysosomes) and enabling unique staining. The LysoTracker probe is effective at nanomolar concentrations, highly selective for acidic organelles, and provides a simple one-step staining method without the need for antibody detection.
Lysosomes and Their Functions
Lysosomes are acidic due to the production and storage of digestive enzymes (hydrolases). This acidic environment is capable of degrading carbohydrates, lipids, nucleic acids, and peptides. Extracellular proteins, viruses, or bacteria can be internalized and transported to lysosomes for degradation via the lysosomal proteolytic pathway. Additionally, misfolded proteins or damaged organelles can become targets for lysosomal degradation through the induction of autophagy. Autophagy is crucial for cell differentiation, survival during nutrient deprivation, and normal growth control. Various pH-sensitive probes can take advantage of the inherent acidity of lysosomes, as well as any changes in pH during biosynthesis or disease processes. LysoTracker products can be used for fluorescent detection of acidic environments in live cells and for labeling and tracking acidic organelles such as lysosomes.
