Background
Mouse IFN-γ, or Interferon-gamma, is a crucial cytokine primarily produced by natural killer (NK) cells and T lymphocytes, particularly Th1 cells and cytotoxic T cells. It plays a pivotal role in the immune response, acting as a key mediator in both innate and adaptive immunity. IFN-γ is known for its potent antiviral, immunoregulatory, and anti-tumor properties. It enhances the presentation of antigens by upregulating major histocompatibility complex (MHC) class I and II molecules on antigen-presenting cells, thereby facilitating a more robust immune response. Stimulate 2×10⁵ cells/well of mouse spleen cells with Con A for 20 hours at 37 °C in a 5% CO₂ incubator. Repeat across 8 wells in 3 batches to evaluate inter-batch and intra-batch precision. Data presented include mean spot count, range, and coefficient of variation.

Stimulate 2×10⁵ cells/well of mouse spleen cells with Con A. After 20 hours of stimulation at 37 °C in a 5% CO₂ incubator, incubate with HRP-labeled detection antibody at room temperature for 2 hours. Add substrate for color development, followed by image analysis and spot counting.
