Background
Human free PSA refers to the portion of prostate-specific antigen that circulates in the bloodstream unbound to other proteins. It is one of several molecular forms of PSA that can be measured through a blood test. The term "free" distinguishes it from "complexed PSA," which is bound to proteins like alpha-1-antichymotrypsin. Clinically, the measurement of free PSA is not used alone but in relation to the total PSA level. This calculation yields the free PSA percentage, which is a valuable diagnostic ratio. A lower free PSA percentage is generally associated with a higher probability of prostate cancer. Conversely, a higher percentage is more often linked to benign conditions, such as an enlarged prostate. Therefore, this test is primarily used to help doctors decide on the need for a prostate biopsy in men with a moderately elevated total PSA.

Linearity
The concentrations of Human free PSA were measured and interpolated from the target standard curves and corrected for sample dilution.
#1 sample is undiluted samples are as follows: Human serum was 2 fold dilution (50%). The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be 442.70 pg/mL in human serum.
#2 sample is undiluted samples are as follows: Monkey serum was 2 fold dilution (50%). The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be 403.32 pg/mL in monkey serum.
Linearity
The concentrations of Human free PSA were measured and interpolated from the target standard curves and corrected for sample dilution.
Sample is undiluted samples are as follows: Human plasma was 2 fold dilution (50%). The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be 1009.32 pg/mL in human plasma.
To assess the linearity of the assay, sample was spiked with high concentrations of Human PSA in various matrices and diluted with the appropriate Calibrator Diluent to produce samples with values within the dynamic range of the assay.
To assess the linearity of the assay, two samples were spiked with high concentrations of Human PSA in various matrices and diluted with the appropriate Calibrator Diluent to produce samples with values within the dynamic range of the assay.
