Background

MUC5B, encoded on chromosome 11p15.5, is a 5,700-amino-acid gel-forming mucin produced chiefly by mucosal epithelial cells in the respiratory tract, gastrointestinal tract, cervix, and salivary glands; it polymerizes via N-terminal von Willebrand factor-like D1-D2-D′-D3 domains that disulfide-link into massive, highly glycosylated (O-linked) linear chains whose extensive carbohydrate content (up to 80 % of mass) endows viscoelastic, lubricating, and antimicrobial barrier properties essential for mucociliary clearance and host defense.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human colon. Anti-MUC5B antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Negative control: IHC shows negative staining in paraffin-embedded human cerebral cortex.

Negative control: IHC shows negative staining in paraffin-embedded human skeletal muscle.

Immunofluorescence

IF shows positive staining in paraffin-embedded human colon. Anti- MUC5B antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. Counterstained with DAPI (Blue). Heat mediated antigen retrieval with EDTA buffer pH9.0 was performed before commencing with IF staining protocol.
