IHC shows positive staining in paraffin-embedded human stomach. Anti-ATP4B antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Background
ATP4B (ATPase H+/K+ transporting subunit beta) is a 291-amino-acid, heavily glycosylated transmembrane protein that forms the regulatory β-subunit of the heterodimeric gastric H+/K+-ATPase proton pump, which exchanges cytoplasmic H+ for extracellular K+ across the apical membrane of parietal cells to drive gastric acid secretion; it stabilizes and targets the catalytic α-subunit to the membrane, acts as a conformational ratchet that locks the pump in its outward-facing E2 state, and contributes to epithelial cell polarity and adhesion through an immunoglobulin-like C-terminal ectodomain .

Immunohistochemistry
Negative control: IHC shows negative staining in paraffin-embedded human gastric cancer.
Immunofluorescence
IF shows positive staining in paraffin-embedded human stomach. Anti-ATP4B antibody was used at 1/250 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. Counterstained with DAPI (Blue). Heat mediated antigen retrieval with EDTA buffer pH9.0 was performed before commencing with IF staining protocol.
