Background

This product is a trypsin analogue derived from microorganisms, the recognition site is lysine or arginine, and the cutting site is the carboxyl end of arginine or arginine residues. It does not contain any animal-derived ingredients, has no animal-derived virus contamination (such as swine influenza virus, porcine parvovirus, etc.), lacks chymotrypsin activity naturally, and is a substitute for pig or bovine trypsin. This product has mild action on digestion cells and is stable and easy to store.

Protocol

Instructions for Use (for reference only)
1. Preheat the recombinant trypsin LS and complete medium at 37℃ before use.
2. Discard the supernatant of the culture medium, wash the cells with an appropriate amount of DPBS, and discard the DPBS.
3. Add an appropriate amount of Recombinant Porcine Trypsin LS (0.5 ml/10 cm2) to the cells and incubate at 37°C until the cells detach.
4. Add 5-10 ml of preheated complete culture medium, gently mix the cells, and transfer the cell suspension to a sterile 15 ml centrifuge tube. Centrifuge at 100g for 5-10 minutes.
5. Discard the supernatant, and add an appropriate amount of complete culture medium for cultivation.

Bioactivity

Normal CHO-K1 cells were digested with imported brand pancreatic enzyme analogue (1×) and UA brand 0.025mg/ml recombinant pancreatic enzyme LS, respectively. ① Cell viability was calculated after digestion at 37℃ for 5min. ② After the cells were digested by pancreatic enzyme, the cell yield was calculated by using pancreatic enzyme digestion. ③ The digested cells were inoculated into 96-well plates and cultured to a certain density to detect cell proliferation activity.

The normal cultured mesenchymal stem cells were digested with imported brand (1×) and recombinant trypsin LS, respectively, and the live cell concentration, cell viability and clumping rate were calculated.
