Background
Mouse Interleukin-5 (IL-5) is a Th2-derived cytokine critical for eosinophil biology. Produced primarily by activated T helper 2 cells and group 2 innate lymphoid cells (ILC2s) in mice, it signals through a heterodimeric receptor (IL-5Rα/βc) to regulate eosinophil differentiation, survival, and activation. This 45-kDa homodimeric glycoprotein drives allergic inflammation and anti-helminth immunity by promoting eosinophil migration to tissues and stimulating immunoglobulin production in B-1 cells. Its pivotal role in eosinophil-mediated pathologies makes it a key therapeutic target in murine asthma models and parasitic infections.

The concentrations of Mouse IL-5 were measured and interpolated from the target standard curves and corrected for sample dilution.
The sample is undiluted samples are as follows: EL-4 cells stimulated with 10ug/ml PHA and 10ug/ml PMA for 2days (5%). The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be 4497.53 pg/mL in stimulated EL-4 cells supernatant.
The sample is undiluted samples are as follows: EL-4 cells culture for 2days (100%). The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be 87.39 pg/mL in unstimulated EL-4 supernatant.
To assess the linearity of the assay, The sample was spiked with high concentrations of Mouse IL-5 in various matrices and diluted with the appropriate Calibrator Diluent to produce samples with values within the dynamic range of the assay.
