Background

Mouse IgG3 is a unique immunoglobulin subclass with several distinctive properties. Structurally, it has an extended hinge region that confers flexibility but also makes it more susceptible to proteolytic cleavage. A defining feature of IgG3 is its exceptional ability to activate the classical pathway of the complement system, often considered the most potent among all mouse IgG subclasses. However, it binds with very low affinity to most Fc gamma receptors (FcγRs), making it relatively ineffective at FcγR-mediated effector functions like ADCC. IgG3 responses are often prominent against polysaccharide antigens, such as those found in bacterial capsules. It also has a tendency to self-aggregate and form cryoglobulins, especially in certain immune complexes.

Standard curve

Linearity
The concentrations of mouse IgG3 were measured and interpolated from the target standard curves and corrected for sample dilution.
#1 sample is undiluted samples are as follows:  CD-1 mouse serum was 2000fold dilution. The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be 83.11 ug/mL in CD-1 mouse serum.
#2 sample is undiluted samples are as follows:  BABL/C mouse serum was 2000fold dilution. The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be 100.29 ug/mL in BABL/C mouse serum.
#3 sample is undiluted samples are as follows:  C57BL/C mouse serum was 2000fold dilution. The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be 129.83 ug/mL in C57BL/C mouse serum.

Linearity
The concentrations of mouse IgG3 were measured and interpolated from the target standard curves and corrected for sample dilution.
#1 sample is undiluted samples are as follows:  C57BL/C mouse plasma (EDTA) was 2000fold dilution. The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be 110.36 ug/mL in C57BL/C mouse plasma (EDTA).
#2 sample is undiluted samples are as follows: C57BL/C mouse plasma (Heparin) was 2000fold dilution. The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be 134.34 ug/mL in C57BL/C mouse plasma (Heparin).
#3 sample is undiluted samples are as follows:  C57BL/C mouse plasma (Sodium citrate) was 4000fold dilution. The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be 76.64 ug/mL in C57BL/C mouse plasma (Sodium citrate).
