Background
Mouse IgG2a is a potent, pro-inflammatory antibody subclass typically induced during Th1-type immune responses, often against viral infections. It is highly effective at engaging effector mechanisms due to its high affinity for specific activating Fc gamma receptors (e.g., FcγRI, FcγRIII, and FcγRIV in mice). This strong binding makes IgG2a exceptionally powerful in mediating effector functions like Antibody-Dependent Cellular Phagocytosis (ADCP) by macrophages, ADCC by natural killer (NK) cells, and robust complement activation via the classical pathway. Because of its superior capacity to recruit and activate innate immune cells, IgG2a is often regarded as the most critical IgG subclass for protective immunity against intracellular pathogens and for the efficacy of many therapeutic monoclonal antibodies. However, a critical point of strain-specific variation exists: the conventional IgG2a isotype is not expressed in the widely used C57BL/6 mouse strain. Due to a natural genetic mutation, C57BL/6 mice produce a functionally analogous but structurally distinct variant called IgG2c instead. Therefore, when evaluating Th1 responses in C57BL/6 mice, assays must specifically detect IgG2c, not IgG2a.

#1 sample is undiluted samples are as follows: BABL/C mouse serum was 8000fold dilution. The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be 42.07 ug/mL in BABL/C mouse serum.
