Background

Thrombomodulin (TM) is a transmembrane glycoprotein located on the luminal surface of endothelial cells in most normal blood vessels that regulates coagulation and inflammation.     Encoded by an intronless gene on chromosome 20, the mature single-chain TM glycoprotein in the human is 557 amino acids long, structurally organized into five distinct regions, including the cytoplasmic and transmembrane domains, a serine/threonine-rich region with an attached chondroitin sulfate moiety, six consecutive EGF-like repeats, and the N-terminal lectin-like domain. Thrombomodulin is an endothelial cell thrombin receptor that converts thrombin into an anticoagulant capable of activating protein C, suggesting a protective role in thrombosis. TM may also affect astrocyte function and protect against thrombin-induced neuronal cell death via an APC-dependent mechanism.
