Background
SKP2 (S-phase kinase-associated protein 2) is an F-box-containing substrate-recognition subunit of the SCFSKP2 E3 ubiquitin ligase complex (Cullin-1-SKP1-SKP2-Rbx1). Its N-terminal F-box docks onto SKP1, while C-terminal leucine-rich repeats (LRRs) capture phosphorylated targets. By catalysing K48-linked poly-ubiquitylation, SKP2 drives proteasomal degradation of a panel of tumour suppressors and cell-cycle brakes, including p27Kip1, p21Cip1, p57Kip2, FOXO1, BRCA2 and RASSF1A, thereby promoting G1/S progression, DNA replication and survival signalling.
