Background
The kit employs homogeneous time-resolved fluorescence resonance energy transfer (TR-FRET) technology to measure the interaction between STAT6 and CRBN mediated by the molecular glue AK-1690 and the compounds to be tested. This method enables a simple, rapid, and high-throughput detection of small molecules that can mediate the interaction between STAT6 and CRBN. As shown, the interaction between STAT6 and CRBN is detected using Eu-labeled anti-Tag1 antibody (TR-FRET donor) and Ac-labeled anti-Tag2 antibody (TR-FRET acceptor). Since the molecular glue AK-1690 mediates the interaction between STAT6 and CRBN, the donor and acceptor antibodies are brought into close proximity. The excitation of the donor antibody triggers fluorescence resonance energy transfer (FRET) to the acceptor antibody, causing the acceptor antibody to specifically emit a signal at a wavelength of 665 nm. This specific signal is proportional to the degree of interaction between AK-1690 and STAT6, CRBN. The homogeneous assay is simple to operate and does not require any washing steps.
