Background

SMARCD1, also known as BAF60A, is a protein that is part of the SWI/SNF family of chromatin remodeling complexes. It plays a crucial role in regulating gene transcription by altering chromatin structure, which involves the alteration of DNA-nucleosome topology in an ATP-dependent manner. SMARCD1 is involved in various biological processes, including neurogenesis and the regulation of lipid metabolism. It is also implicated in diseases such as Coffin-Siris Syndrome and has been identified as a prognostic marker in liver hepatocellular carcinoma. The protein is ubiquitously expressed in the nucleus of various tissues and is essential for the activation of specific gene signatures in multipotent progenitors.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human testis. Anti-SMARCD1 antibody was used at 1/200 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
