Background

Aquaporin 5 (AQP5) is a water channel protein that plays a significant role in the generation of saliva, tears, and pulmonary secretions. It is a member of the major intrinsic protein family, characterized by its ability to selectively transport water across cell membranes. AQP5 is predominantly expressed in the salivary and lacrimal glands, as well as in the respiratory tract, and is crucial for maintaining water homeostasis within these secretions. The protein is also implicated in the pathogenesis of certain diseases, such as Sjögren's syndrome, where its abnormal subcellular distribution in salivary gland cells may contribute to the disease's development.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human testis. Anti-Aquaporin 5/AQP5 antibody was used at 1/500 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunofluorescence

IF shows positive staining in paraffin-embedded mouse lung. Anti- Aquaporin 5/AQP5 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. Counterstained with DAPI (Blue). Heat mediated antigen retrieval with EDTA buffer pH9.0 was performed before commencing with IF staining protocol.
