Background
HER4, also known as ErbB4, is a member of epidermal growth factor receptor (EGFR) or ErbB family. The HER4 gene is localized on chromosome 2q33.3–34. HER4 is necessary for normal development of body systems including heart, nervous system, and mammary gland. HER4 is commonly expressed in many human tissues, both adult and fetal. EGFR/ErbB1/HER1, ErbB2/HER2, ErbB3/HER3, and ErbB4/HER4 are members of this receptor tyrosine kinase family. HER4 is the only member of this family of receptors with growth inhibiting properties. HER4 can activate related genes in the nucleus when combined with epidermal growth factor (EGF), promoting cell division and proliferation. It is activated through ligand binding and activates MAPKs and PI3K/AKT pathways. Her4 promotes glycolysis, glutaminolysis, mitochondrial biogenesis & OXPHOS through c-Myc-dependent signaling axis in cancer. The Her4-mediated metabolic reprogramming may in part cooperate to promote tumorigenesis in certain cancers.
