Background
IKZF1 is a zinc-finger-containing transcription factor belonging to the Ikaros family. It is predominantly expressed in lymphocytes and regulates target gene transcription by recognizing and binding to specific DNA sequences, serving as a critical regulator of hematopoietic and immune system development. It acts as a "master regulator" in lymphocyte differentiation: in the B-cell lineage, it promotes early B-cell development and guides their differentiation into antibody-secreting cells, while also participating in immunoglobulin gene rearrangement; in the T-cell lineage, it controls T-cell maturation, lineage selection, and thymocyte proliferation, while maintaining genomic stability in developing cells. Ikaros functions also extend to the innate immune system, such as regulating the development of dendritic cells and NK cells. Consequently, IKZF1 abnormalities are closely associated with various diseases—its mutations or deletions can lead to hematological malignancies such as B-cell acute lymphoblastic leukemia (B-ALL), while alterations in the IKZF1 gene are also considered risk factors for common variable immunodeficiency (CVID) and autoimmune diseases.
